Cryostat Do's and Don'ts
Working with cryostat machines requires precision and care. Here are essential guidelines I've learned through extensive lab work.
Do's
- Always pre-cool the chamber before beginning work
- Use fresh, sharp blades for clean sections
- Maintain consistent temperature throughout the process
- Handle tissue blocks carefully to avoid damage
- Clean the machine thoroughly after each use
- Document section thickness accurately (20–40µm range)
Don'ts
- Don't rush the cooling process
- Don't use dull blades - they cause artifacts
- Don't skip cleaning steps between samples
- Don't ignore temperature fluctuations
- Don't force tissue that's too hard or too soft
Best Practices
From my experience teaching undergraduates, the most common mistakes come from impatience. Take time to prepare properly, and the sections will be clean and consistent. Quality over speed always.
